Aminopterin (4-aminofolic acid) is an amino derivative of folic acid, an antimetabolite drug with cytostatics effects. It blocks the synthesis of tetrahydrofolate by binding competitively to the dihydrofolate reductase enzyme. Methotrexate has the same effect.
Aminopterin has been proven to have characteristic teratologic effects that can cause severe congenital malformations of the skull, skeleton or anencephaly.[1].
